T-Cell Clonality Assessment by Restriction Fragment-Southern Blot Hybridization, Fluid : 0055596

Mnemonic: TCRFSBHF

Methodology: Restriction Fragment Southern Blot Hybridization
Performed: DNA isolation: Sun-Sat; Assay: Varies
Reported: 7-14 days
Specimen Required: Collect:  Two 5 mL lavender (EDTA) or pink (K2 EDTA), one 10 mL yellow (ACD), or 3 mL bone marrow.

Transport:  10 mL whole blood or 3 mL bone marrow (EDTA) at 2-8°C. (Min: 3 mL)

Pediatric Collection/Transport:  3 mL whole blood or 1 mL bone marrow (EDTA) at 2-8°C.

Unacceptable Conditions:  Serum, frozen whole blood, clotted blood, and severely hemolyzed samples.

Stability:  Ambient: 24 hours; Refrigerated: 5 days; Frozen: Unacceptable
Reference Interval:
Negative: A monoclonal T-cell population is not detected.
Positive: A monoclonal T-cell population is detected.  (Monoclonal T-cell receptor beta rearrangement identified by Southern blot analysis.)
Interpretive Data: By report

Please refer to Statement B in the Compliance Statements section in the front of the Laboratory Test Directory.
Note: A positive result indicates the presence of monoclonal T-cell population within the sample.  A negative result does not entirely exclude the presence of a T-cell receptor gene rearrangement (or monoclonal T-cell population) in the sample.
CPT Code(s): 83891 Isolation; 83892 x3 Digestion; 83894 Gel Electrophoresis; 83897 Southern transfer; 83896 Nucleic acid probe; 83912 Interpretation and report - Additional CPT code modifiers may be required for procedures performed to test for oncologic or inherited disorders.
