Collect: One 10 mL yellow (ACD Solution A) and one normal control in a 10 mL yellow (ACD Solution A) from a healthy unrelated individual.
Transport: 10 mL whole blood and 10 mL normal control in sterile containers at 20-25°C. LIVE CELLS REQUIRED. CRITICAL AMBIENT. Do not refrigerate or freeze. Pediatric Collection/Transport: 3 mL whole blood (green, sodium heparin) and 3 mL whole blood (green, sodium heparin) normal control from a healthy unrelated individual in sterile containers at 20-25°C. Remarks: Patient and control samples must be collected within 48 hours of test performance. Unacceptable Conditions: Refrigerated or frozen samples and samples in transport longer than 48 hours Stability: Ambient: 48 hours; Refrigerated: Unacceptable; Frozen: Unacceptable |

PAM3CSK4, synthetic bacterial lipoprotein (TLR2-TLR1 ligand), Zymosan Cell wall particles from Saccharomyces cerevisiae (TLR6-TLR2 ligand), Poly (I:C), synthetic analog of dsRNA (TLR3 ligand), Lipopolysaccharide Ultra pure S. minnesota LPS (TLR4 ligand), Flagellin, purified from S. thyphimurium (TLR5 ligand), Loxoribine, guanosine analog (TLR7 ligand) are tested independently in peripheral blood mononuclear cell (PBMC) culture. PBMC cytokine production responses to these mitogens are determined by multi-analyte fluorescence detection.
