Specimen Required:  
Collect: One 4 mL green (sodium heparin) AND a normal control in one 4 mL green (sodium heparin) from a healthy unrelated individual. Also acceptable: Yellow (ACD solution A).

Transport: 4 mL whole blood AND 4 mL normal control whole blood in sterile containers at 20-25°C. LIVE LYMPHOCYTES REQUIRED.

Pediatric Collection/Transport: 2 mL whole blood AND 2 mL normal control whole blood in sterile containers at 20-25°C. LIVE LYMPHOCYTES REQUIRED.

Remarks: CRITICAL AMBIENT. Do not refrigerate or freeze. Patient and control specimens must be collected within 48 hours of test performance.

Unacceptable Conditions: Refrigerated or frozen specimens. Specimens in transport longer than 48 hours.

Stability: Ambient: 48 hours; Refrigerated: Unacceptable; Frozen: Unacceptable

Interpretive Data:
Deficiency in cytokine production in response to stimulation suggests possibility of defect in Interleukin-1-Receptor-Associated Kinase-4 (IRAK-4), which may result in recurrent pneumococcal and/or staphylococcal infections, in absence of other abnormalities of host defense.
